Experimental DNA vaccine gives full protection against herpes

Coridon has announced the results of a pre-clinical animal study of new DNA vaccine showing 100% protection against herpes simplex virus 2.

Brisbane-based Coridon has reached another milestone in the development of its pioneering DNA vaccine against herpes simplex virus 2 (HSV-2) with results from a pre-clinical efficacy study showing it was 100 per cent effective in protecting animals against the virus.

Coridon now plans to progress the DNA virus programme into human clinical studies, with funding from its major investor, Allied Healthcare Group (ASX:AHZ).

The vaccine was developed by Professor Ian Frazer and his team at Coridon, and the study was done in collaboration with Professor David Koelle at the University of Washington in Seattle.

The results of the study were presented at the 5th Vaccine and ISV Annual Global Congress in Seattle on 3rd October 2011.

“The results of our herpes vaccine mark the beginning of an exciting period,” said Professor Frazer.

“Over the next 12 months, we expect pivotal data showing that our HSV vaccine, which incorporates Coridon optimisation technology, produces similar immune responses in the clinic to those seen in the animal trials.”

The DNA vaccine works differently to conventional vaccines. DNA from the virus is injected into the individual, and the body generates an immune response against the proteins encoded by that DNA.

This then enables the body to mount an effective immune defence against the full virus should it ever appear.

DNA vaccines have the tremendous benefit of not only offering prophylactic protection, but also potentially curing individuals currently infected by the virus.

Coridon is developing DNA vaccines for the prevention and treatment of a range of infectious diseases and cancers in humans.

I sincerely hope that the human clinical trials are 100% successful like the animal trials, and that the vaccine has therapeutic/curing abilities as well as protective abilities. If so, this is the miracle drug that we all have been waiting for and if they host clinical trials in the US, I'll be the first in line for this one! Hope they keep us posted!

I would be keen on a HSV-1 cold sore vaccine, which I understand is very closely related to HSV-2. I have always been terrified of contracting cold sores considering how prevalent they are and many people don't care about spreading it.

If cold sores could be cured with the same or similar vaccine, then that would potentially be a much larger market than just HSV-2.

This is amazing news, especially for people suffering with HSV-2. There are more people living with HSV-2 than HSV-1. Cold sores are nothing compared to the awful blisters you get from HSV-2. Please bring this to the states soon!!! We need to cure this awful disease!!

I would also desperately love a HSV-1 vaccine. People tend to think of coldsores as minor insignificant things but if you are immunosuppressed they are scary, they can cause blindness etc etc and wreak havoc with your body.we sorely need a HSV-1 vaccine, I hope this technology can easy move their once this is done

I have been infected wih HSV-2 a year ago and ever since I get very depressed when I think about it and how I can move on and enjoy a healthy family life with this disease. With this news, I can only pray this works in clinical trials and gets to the market.

At this point, I don't even want a cure! Just a vaccine to stop scaring people away!

I just got diganosed the other day, still trying to get my head around it.

How long does it usually take, from animal testing to the actual vaccine to be available? A year? 2?

With more than 500 million people infected worldwide, this vaccine will be a money generator. Then in the US alone, approximately 2,200 new people are infected with hsv2 daily. Antivirals are becomig absolute and us who suffer from ineffective treatment, also suffer from painful outbreaks, severe depression, isolation, and have feelings of hopelessness. The market for herpes treatment is estimated at $1 billion. Too many people are becoming infected and spreading the virus because they dont know they are infected. Then for people like myself suffer greatly because current antivirals are ineffective. My outbreaks are literally nonstop and my love life is ruined. Having children is only a dream. My relationship with the love of my life is coming to an end. All over a virus I have no control over.

A vaccine that can suppress HSV (1-2) is worth billions. From what I read (CDC/Medical Journals) this virus is quite prevalent. So much so that upwards of 40% of the American population - similar in most countries - have this in there system. Since most people dont realize they have it, it spreads. This is more prevalent than HIV, it also facilitates HIV. It seems as though it's a pandemic which is not spoke about.

I am hoping that you can share these stories with Professor Ian Frazer and his team. These are only a few stories from people suffering from HSV - the reality is that there are millions of people around the world afflicted with this virus. HSV testing is also not included in the standard panel of STD tests so many people are not aware that they have HSV as they are asymptomatic but they can easily pass on this virus to others. We URGENTLY need a therapeutic vaccine that can suppress the virus so that it is not passed on to others. Please Professor Frazer help us. Thank you.

This s**t is ruining my love life I would not even dare to kiss
someone anymore from fear of spreading my nightmare.
I would pay for other researches too since I now understand how if feels to have something against your own will!
This has completely change my view on life. I once was pretty more superficial than I am.
Now I'm constantly fighting against superficiality. It kinda kills any good vibe and this is a problem you live alone since it's considered in our society as a real shame !
The visual factor in the disease is just the tip of the iceberg,the worst is when groups of people makes joke about that but they have no idea that one of their best friend has that since he was young (me) the psychological effect is disastrous no wonder it's been related to alzeimer!

I'm so thrilled that finally after decades there is something good coming out in the field of herpes treatments. It's more than a dream come true because this treatment if completely successful can give me my old life back and millions of others. I miss the spontenous, spunky, eager, and oooh so funny Corina I use to be. I'm an educated individual that pays my taxes and serves my country well. To live with one of the most stigmatized diseases known to man is as bad as having cancer. Mental health is highly important and to feel like you're trapped in a prison because of a painful virus is debilitating. I hope within the next decade, maybe a couple years sooner, this vaccine is made available for human consumption through a doctors approval. Herpes has been put on the back burner for far too many years and the suffering is unbearable. Now, the demand for superior herpes treatment is here, you just need to create the supply!!!

I would like to take the time to thank Coridon and Prof. Frazer for their research in the areas of therapeutic and prophalyphic HSV vaccines and treatments. HSV is a silent disease/infection that is often unknowingly spread to others. I, myself, didn't realize how serious of an epidemic HSV was until recently infected.

My life has changed drastically since the date of my diagnosis last month. The psychological effects of HSV greatly decreases the quality of life of those living with the disease, and I can't imagine living the rest of my life with this stigmatized and widely misunderstood virus.

There are millions of sufferers worldwide that are in desperate need of better treatment options. Although viral suppression does lend some control and sense of normalcy to sufferers, complete eradication/ cure would be ideal and absoultely AMAZING ! You all have plenty of supporters who are anxiously waiting for your vaccine's arrival to the market.

I was diagnosed with HSV 2 five months ago and I was and still am devasted by the diagnosis. Life as I once knew it no longer exists for me. I am having back to back painful outbreaks and I am now also suffering from depression. Sigh...Words can't express the gratitude I feel to know SOMEONE is working on a cure(more research is needed). I'm actually a prospective medical student and I plan on doing my own research during/after med-school to find a cure... I guess what hurts the most is the fact that I didn't sleep around. HSV needs a cure and the work you are doing has given hope. While HSV is not considered as a serious health problem by the medical community I assure you it is by those who suffer from it. Additionally, new research is showing that the virus (especially HSV-1) has been linked to Alzheimer's and is perhaps more dangerous then thought- I am actually asymptomatic and would love a cure--it would give me my life (i know this sounds dramatic to someone without HSV)! Ignorance lead to me contracting this virus and I have had to pay a very heavy price for it, i'm with a woman who loves me very much but we are not able to express that love physically.
We want to get married but there is the concern of how we would go about starting a family, where we live medical assistance in aiding conception of a child is extremely expensive and I do so want a child with her.
I see alot of research going on for HSV cures and permanent suppressive therapy and i am hopeful there will be a break through soon that will offer a real long term solution to HSV sufferers like myself. When that day comes I will propose to her.

I was diagnosed with herpes about 9 years ago. I have noticed this horrible virus gets worse every break out. When I say the break out gets worse Im not reffering to the location of the breakout, this I have been able to keep contained to the initial breakout area. I am reffering to the symptoms that come with it. I suffer from chronic migraines almost on a daily basis, I become very irritable, emotional, Im always extremely tired and the list goes on and on. I was also diagnosed with fibromyalgia at the age of 28 with no tramatic experiences happening in my life,I cant help wonder if herpes carries a role in this. Im so horribly afraid of giving this to my kids thru the simplest of things, using my towel, taking a bath after me, using my soap. Its a horrible feeling. I just wanted to say Thank You for hopes of a near cure!

This is a psychology and mentally depressing virus. I would love to have meaning in my life again after 1 1/2 years of mental anguish. The depression everyday is slowing killing me with thoughts of hopelessness and not feeling worthy or human...almost like having leprosy Serioulsy this is how this has made me feel and from what I've read others. Please bring us normacy again. When most of the thoughts in your day no matter at work, working out, cooking for your kids, being with friends etc. is on this virus....that is a sad life. I've had HSV2 for 6 and a half years. Although I've gotten to an acceptance stage, I've still not had a relationship. I think this is due to not meeting the right person, not the Herpes.

I've finally met someone I want to be with and although we have not started to sleep together, I'm paranoid about giving it to him. This makes life hard.

If this vaccine does work on us humans, it would give piece of mind to thousands/millions of people.

My doctor tried to comfort me by saying that this is not the worst thing in the world, but only someone knows how uncomfortable knowing you have a disease that has no cure.
live the fear of infecting other people and make them go through what I did.
since I discovered to be the bearer of this bad, I'm not the same I'm afraid I relate and I try not to comment on this subject for fear of being discriminated.

Excellent!! Hsv can really change lives and not necessarily for the better. Those who suffer outbreaks periodically feel especially isolated and usually in trouble to start new relationships or not to spread it to a hsv negative partner. I pray to god to support you in your research. With your new vaccine, u can save millions of sufferers. Herpes is scary cos you can pass on unknowingly. If we can only have vaccine to protect those who are negative so be it. Just like hep b vaccine. At least our partner won't leave us because of this. I have lived with HSV for 5 years and recently I transmitted the disease to my genitalia and it is now on my arms (from bathing). I have 2 children (7 and 11) that I am not hugging for fear that I may infect them. Bless you for working on a vaccine and cure.

This is wonderful news and I would like to pass on my thanks and congratulations to Professor Frazer and his team. I was diagnosed with HSV2 13 years ago and in the past 18 months I have suffered contant symptoms and outbreaks. It is absolutely depressing and keeping me from finding a loving relationship as I can't imagine passing this on to anyone! It has been a very hard thing for me to live with as I have always been very healthy. The anti-virals on the market now do nothing for me. It would change my life to see this drug on the market! I hope and pray it doesn't take long.

I cant begin to thank you for your efforts,..and diligent efforts in helping all of our poor souls who have been stricken with this debilitating disease. some say its not so bad!..Well I say it is! I gave it to my wife and my two children as I never knew I was infected. They did nothing to deserve this, I'm in the restaurant buis. and had to quit my job as the outbreaks were so bad, I could no longer perform the daily task and not be scrutinized. Its the kids I feel the worst for, the other kids can be so cruel, and the ridicule is so terrible for them at school. it breaks my heart,..to know I've done this to my Family. Please continue your quest, as so,so,so many of us thank you for your efforts.

This is the first time i have spoken out. A glimmer of hope has
appeared .

This relentless disease has harassed me for over five years now.
My constant battle to put it (hsv2) at bay is proving very difficult .
In the meantime hsv2 has taken my income , my companionship
and now it seems depression is becoming part of the battle.

I extend my highest graditude to Professor Frazer and the team at Coridon for devoting there time and effort to eradicating this disease from the people.

Sounds great and more promising than other trials I have read about. Has there been any update since this publication as I am eager to find out more. I don't want to get my hopes up only to see dem dashed once again by unsuccessful trials. Every few years u read about how close science is to finding a cure and then in the end there's nothing.

Thank you so much for all of your wonderful efforts and advances in finding a cure for HSV. I live in Melbourne and have just been diagnosed with HSV1. The news has devastated me especially since i've led such a careful life. Now my days are not so care-free anymore as the topic of the Herpes virus and how to treat it constantly plagues my mind. But reading this has given me so much hope. I would be absolutely overjoyed and incredibly grateful if you found a cure for HSV 1 & 2...and i'm sure millions of people would be too. I know you have heard it many times in these posts but THANK YOU for your hard work in finding a cure. So many people's lives will be changed for the better once this is successful including my own.

The day I found out I had Herpes I thought my life was over. I thought I could never have a normal life and have been trying to do so. However everyday I am reminded of what I have and that it supposedly will be with me forever.

If you need someone for clinical testing, I would definitely volunteer. Don't know if you will see this post but Thanks again and Good Luck!

[My apologies for the delay in replying. Thank you for your enquiry about our clinical trials program.

These are expected to start this year in Brisbane, and when the studies are open for recruitment, information will be posted on our website www.coridon.com.

The first study will be seeking uninfected volunteers.

We do not envisage conducting trials in the US for a couple of years.

I am a 32 year old woman and I have been living with hsv type 2 for 8 years . It changed my life so much. I used to love to work out and look pretty but once I was diagnosed I fell in to a huge depression. I gained weight and lost my self esteem. I often have painfull outbreaks that effect my ability to exercise, and often effect my ability to preform my job as well as I would like. I even decided to not have another child due to this disease. I would be extremely greatfull if my husband and I could be considered for your clinical trials when you start doing human trials. I look forward to the day that people like me can live a normal healthy active life and not feel the weight of the stigma this disease has place on our shoulders. Thank you so much for looking at this disease like it is a real health issue and I pray that all of your research is speedy and sucsessfull.

I would take out a loan on my house to pay for a treatment to cure this Disease. I have had it 5 years due to my unfaithful wife and it has been absolute hell. Side affects of current medical treatments are hardwork also. I have tried Chlorine treatment, 20,000mg injections of Vit C , Liquid Zeolite , Urine therapy, Chinese medicine , Homeopathy all of the Natural therapies.... None work. Hi, This is great news . I definitely think there should be a vaccine for the hsv1 for cold sores on mouth as this is the stepping stone to to spread in the genital parts . 90% of people have it. For people who are serious about each other and marry at a later age in life the spreading starts. Hsv1 for infected and uninfected. Hsv2 vaccine can this be work for hsv1.
